If you are behind your intended victim and wish to kill them quickly with a knife, the best way is to stab the side of the neck and slash your way out of the neck, opening up a significant portion of the neck. If you are in front, stab them as many times as necessary in the vital organs. If you want to use your bare hands, simply apply any choke that cuts off both carotid arteries, and after they pass out you can heel kick them in the kidneys as much as you want (until they wake up, in which case stomp on their head until their skull caves in).
